Catapult Pharmacology
Platinum-based Therapies
Carboplatin, cisplatin, and oxaliplatin are platinum-based compounds that bind to chloride when in plasma but release the platinum intracellularly, which allows the drug to bind to DNA. Binding to DNA limits DNA replication and synthesis of RNA. Platinum-based drugs can cause neurotoxicity, nephrotoxicity, and myelosuppression.
Class Drugs
Carboplatin
Cisplatin
Oxaliplatin
Class Mechanism of Action
Bound to chloride in the plasma, but loses chloride intracellularly, which allows the drug to bind to DNA
- DNA replication and RNA synthesis is inhibited
Class Applications
See individual drugs
Class Adverse Effects
Ototoxicity
Nephrotoxicity
Myelosuppression
Hypersensitivity reaction
