Denosumab
Denosumab is a monoclonal antibody treatment for osteoporosis and works by inhibiting osteoclast formation and function. Denosumab inhibits osteoclasts by binding RANKL (nuclear factor kappa-B ligand), which is an activator of RANK. RANK is a receptor on osteoclasts and osteoclast precursors that, when activated, promotes osteoclast formation, function, and survival. By inhibiting RANKL, osteoclast activity is reduced, which improves bone density. In addition to osteoclast inhibitor activity that results in risk of osteonecrosis of the jaw and hypocalcemia, monoclonal antibodies also increase the risk of infection.
